LIM1215 cells: Casting light on metastatic colorectal cancer

This year we will be highlighting some of the cell lines from our collection, to see how they are contributing to research discoveries and clinical translation. LIM1215 is one of our most popular cell lines and was originally deposited in our collection by the Ludwig Institute for Cancer Research in Melbourne. The cell line was established from a 34-year-old male with hereditary non-polyposis colorectal cancer (HNPCC or Lynch syndrome). LIM1215 cells grow quickly and can be cloned at low density, making them relatively easy to work with [1].
These cells express normal EGFR levels and are sensitive to cetuximab, an anti-EGFR antibody that is used as targeted therapy for metastatic colorectal cancer. If LIM1215 cells are continuously exposed to cetuximab they become resistant to it, due to the emergence of KRAS and EGFR mutations [2-3].
These discoveries mean that LIM1215 has become a valuable model for metastatic colorectal cancer. Patients who receive cetuximab often experience an initial response before becoming resistant to the drug. LIM1215 cells have been used to discover the mechanisms behind this secondary drug resistance and test combination therapies to potentially overcome it [2-4].
LIM1215 also shows promise as a model for new technologies. For example, extracellular vesicles from LIM1215 cells have been isolated, loaded with doxorubicin, coated with a colorectal cancer-specific antibody, and used as a targeted form of therapy in vitro [5-6].
Forty years after it was established, LIM1215 continues to have an important role in developing better diagnostic tools and treatments for a disease where very few options are available.
